Go to your Desktop, click on My Computer, locate your floppy drive.

This appears to have nothing to do with the database software Microsoft Access, the subject of this newsgroup. But to delete a folder from a 3.5 disk simply open the A: drive (or whatever drive letter contains the disk) in Windows Explorer or My Computer, select the folder, and hit the Delete key on your keyboard.
